DEATHLOOP puts players in the role of Colt, an assassin stuck in a time loop on Blackreef island who must kill eight targets known as Visionaries in a single day to break the loop. The island is split into four areas that can be visited at four times of day: morning, noon, afternoon, and evening. Players use guns, melee attacks, and supernatural powers to fight enemies. These powers include teleporting short distances, moving objects with your mind, and turning invisible. If Colt dies, the day restarts, and he keeps his memory but loses weapons unless a special resource called Residuum is used to save them. Another assassin named Julianna hunts Colt throughout the game, controlled by AI or a real player in online mode.

The Deluxe Edition includes unique weapons, character skins, and two trinkets that give special bonuses. The game lets players approach goals in any order and style, using stealth or direct attacks. Information learned in one loop can be used in future attempts, such as door codes and target locations. Players gain powers from slabs carried by Visionaries, which can be upgraded by killing the same Visionary again in a different loop. The island changes based on the time of day, with different events and character locations. The story slowly reveals why the loop exists and Colt’s connection to it through found notes, audio logs, and conversations.
